زينة زامل تركي الغزيأ.م.د طالب فالح حسنأ . سالم حسين محمد2025-05-192025-05-192008-06-25https://dspace.utq.edu.iq/handle/123456789/1003A total of six-hundred and forty-seven samples were collected from patients suffering from different infectious diseases whom were admitted in Al-Imam Hussein educational hospital in Nasiriyah province, during the period from 1/11/2006 to 1/4/2007. These samples were included swabs from (burns, wounds, ears, skin, eyes and vaginal) and samples from (blood, urine, sputum, seminal fluid, pleural fluid and cerebrospinal fluid). Diagnostic results showed that 248 isolates were related to Staphylococcus Spp. and 102 isolates were related to Staphylococcus aureus. Sensitivity test of these isolates to the antibiotics showed higher resistance to Cefotaxime , Amoxicillin, Cephalexin , Cephalothin , with the proportion of ( 94.87% , 92.31% , 87.18 % , 79.49% ) respectively in addition to moderate resistance the Gentamycin , Tobramycin , Ciprofloxacin, Norfloxacin , Trimethoprim with the proportion of ( 47.44%, 52.56%, 42.31%, 42.31%, 52.56% ) respectively.While the isolates were resist to Rifampicin , Chloramphenical , Nitrofurantoin with the proportion of (33.33% , 34.62% , 35.9%) respectively. The isolates that showed resistance to more than 50% of the antibiotics (39) isolates, were tested to produce β-lactmase enzyme, all these isolates gave a positive results to β-lactmase production with an absolute proportion of 100 %. Two types of blood, human (AB) group and sheep blood were used to detect the ability of these isolates to produce hemolysin. It is found that the isolates gave best hemolytic results by using sheep blood with hemolytic zone more than 3mm. 78 isolates (76.47%) showed their ability to produce hemolysin .The hemolytic activity and protein concentration for five isolates were measured, therefore the isolate that gave the highest hemolytic activity (3.51u/mg) and the highest protein concentration (13.43mg/ml) was chosen in the process of hemolysin extraction. After extraction and purification of hemolysin the results showed that the enzyme wouldn’t affect by the purification steps.The results of gel filtration showed that the enzyme has one peak. Activity of the purified hemolysin is tested on the human white blood cells which found that hemolysin has lethal effect on it with concentration 20 µg/ml during 60 minutes. Also it was found that hemolysin has lethal effect when it was injected on a group of laboratory animals (mice) with concentration 22 mg / kg . جمعت 647 عينــة من المرضى الراقديــن والمراجعيــن لمستشفى الإمــام الحسين التعليمي في محافظة ذي قار والذين يعانون من حـالات مرضية مختلفة وللفترة من 1/11/2006 ولغاية 1/4/2007 شملت مسحات ( حروق ,وجروح ,وأذن ,وجلدية ,وعيون ,ومهبلية ) وعينات (دم ,وإدرار,وقشع ,وسائل منوي ,وسائل الجنب ,وسائل النخاع الشوكي ) أظهرت نتائج التشخيص أن 248 عينة تعود لجنسStaphylococcus منها 102 عينة تعود للنوع Staphylococcus aureus . اختبــرت حساسية العزلات المنتجة للهيمولايسين لـ 12 مضادا حياتيا ، وجد إن العزلات كانت عالية المقاومة للمضادات الحياتية Cefotaxime, Amoxicillin , Cephalexin , Cephalothin وبنسبة ( %79.49, %87.18, % 92.31,% 94.87)على التــوالي , بينـما أــدت العزلات مقاومة متوسطة للمضـــادات الحياتيـــةGentamycin , Tobramycin , Ciprofloxacin ,Norfloxacin ,Trimethoprim وبنسبة (47.44 % ,52.56 % ,42.31 % ,42.31 % ,52.56 %) على التوالي.فيمـــا كانت نسبة مقاومة العزلات للمضادات الحياتيـة Rifampicin , Chloramphenicol , Nitrofurantoin هي(33.33% (%35.90, %34.62 , على التوالي. تم الكشف عن قابلية العزلات المقاومة لأكثر من 50% من المضادات الحياتية المستخدمة في الدراسة وكانت 39 عزلة على إنتاج إنزيم البيتالاكتاميز ، فوجد إن جميع العزلات كانت منتجة له وبنسبة 100%. استخدم نوعان من الدم هما دم الإنسان صنف AB ودم الأغنام للكشف الأولي عن قدرة عزلات S.aureus على إنتاج الهيمولايسين فوجد إن العزلات أعطت نتائج تحلل أفضل باستخدام دم الأغنام وبقطر تحلل 3 ملم فأكثر. درست قابلية عزلات S.aureus على إنتاج إنزيم الهيمولايسين فوجد إن 78عزلة76.47) %) كانت منتجة لهذا الإنزيم . تم قياس الفعالية التحللية وقدرتركيزالبروتين لخمس عزلات أعطت أكبر منطقة تحلل على وسط أكار الدم وعليه تم اختيار العزلة التي أعطت فعالية عالية وكانت 3.51 وحدة/ملغم وتركيز بروتين يساوي 13.43 ملغم/مل في عملية استخلاص إنزيم الهيمولايسين. تم استخلاص وتنقية إنزيم الهيمولايسين ، ووجد عدم تأثر فعالية الإنزيم بجميع مراحل التنقية ، وظهر من خلال نتائج الترشيح الهلامي بأن الإنزيم متكون من قمة واحدة. اختبرت فعاليــة الهيمولايسين المنقى على خلايـا الدم البيض للإنسان فوجد بأن للهيمولايسين تأثيــرا قاتلا لها وبتركيز 20 مكغم / مل وخلال 60 دقيقة. كما وجـد بأن للهيمولايسين تأثيــرا قاتــلا عند حقنــه في مجموعة من الفئران المختبرية وبتركيز 22 ملغم / كغم.text::thesis::master thesis
